ABSTRACT

Objective:To investigate the clinical factors influencing the efficacy and prognosis of neoadjuvant chemoradiotherapy (nCRT) in patients with locally advanced rectal cancer (LARC).Methods:The clinical data of 335 patients with LARC who underwent radical operation after nCRT in Liaoning Cancer Hospital were retrospectively analyzed.Through univariate and multivariate analysis, we found out the clinical factors that may affect the efficacy and prognosis of nCRT in patients with LARC.Results:Multivariate logistic regression analysis showed that the independent clinical factors influencing the curative effect of nCRT in LARC patients were tumor invasion circumference( OR=2.350, 95% CI: 1.438-3.842, P=0.01), cT stage( OR=2.101, 95% CI: 1.024-4.314, P=0.043) and cN stage( OR=5.836, 95% CI: 3.305-10.306, P<0.01). Competitive risk model analysis showed that the independent factors affecting the prognosis of LARC patients were tumor regression grading (TRG) ( HR=3.236, 95% CI: 1.714-6.110, P<0.01), cT stage ( HR=1.852, 95% CI: 1.061-3.230, P=0.030), cN stage ( HR=2.008, 95% CI: 1.083-0.372, P=0.027), and carcinoembryonic antigen (CEA) level before nCRT ( HR=7.038, P<0.01), 95% CI: 3.894-12.720, P<0.01) and serum carbohydrate antigen 199 (CA199) ( HR=3.849, 95% CI: 2.016-7.350, P<0.01). Conclusion:The larger the circumference of tumor invasion before nCRT, cT stage and the cN stage were independent clinical factors influencing the nCRT efficacy of LARC patients. The larger the degree of tumor invasion to the rectum, the higher cT stage and the higher the cN stage before nCRT, the worse the efficacy of nCRT in LARC patients may be. TRG grade, cT stage, cN stage, CEA and CA199 level before nCRT were independent factors affecting the prognosis of LARC patients who received nCRT. The higher the TRG grade, of LARC patients receiving nCRT, the higher cT and cN stage, and the higher the CEA and CA199 level before nCRT, the worse the prognosis of LARC patients may be.

The ratio of abnormal perfusion in PLD=2525ms group[CBF(34.81±11.68)mL·100g-1·min-1]was higher than that in PLD=1 525ms group[CBF(18.33±6.42)mL·100g-1·min-1], the difference was statistically significant(Z=-9.196, P<0.001). The percentage of abnormal perfusion in PLD group of 2 525ms(56.2%) was lower than that in PLD group of 1 525ms(94.5%), the difference was statistically significant(χ2=5.393, P=0.02).@*Conclusion@#Dual-phase ASL(PLD=1 525ms, 2 525ms) combined with ASL can more accurately evaluate the compensations of fast collateral circulation and slow collateral circulation, and provide individualized treatment.

ABSTRACT

BACKGROUND:Hydroxyapatite/poly-L-lactic acid composite can wel improve the brittleness and mechanical properties of the hydroxyapatite. OBJECTIVE:To study the biocompatibility of hydroxyapatite/poly-L-lactic acid composite biomaterials with periodontal ligament cels. METHODS:Passage 4 human periodontal ligament cels at a density of 5×109/L were cultured with hydroxyapatite/poly-L-lactic acid composite as experimental group, and human periodontal ligament cels cultured alone as control group. Cel proliferation was detected within 7 days of culture; alkaline phosphatase activity was detected at 24, 48 and 72 hours of culture; and type I colagen expression was detected at 72 hours of culture. RESULTS AND CONCLUSION:There was no significant difference in the absorbance value of cels between the two groups at 1-7 days of culture. The alkaline phosphatase activity of cels at 48 and 72 hours was significantly higher than that at 24 hours (P < 0.05) in the two groups, but there was no difference between the two groups. The expression of type I colagen had no difference between the two groups. These findings indicate that the hydroxyapatite/poly-L-lactic acid composite biomaterial has good cel compatibility.

ABSTRACT

OBJECTIVETo investigate the protective effect of hydrogen against hyperoxia-induced oxidative stress injury in premature rat type II alveolar epithelial cells (AECs).

METHODSThe type II AECs isolated from premature rats were randomly divided into air (21% oxygen) control group, hyperoxia (95% oxygen) control group, air + hydrogen group, and hyperoxia+ hydrogen group. The cells with hydrogen treatment were cultured in the presence of rich hydrogen. After the corresponding exposure for 24 h, the cell morphology was observed microscopically. MTT assay was used to evaluated the cell proliferation ability, and JC-1 fluorescence probe was used to detect the mitochondrial membrane potential (δφ) changes of the type II AECs. The concentration of maleic dialdehyde (MDA) and superoxide dismutase (SOD) activity in the cell supernatant were detected using colorimetric method.

RESULTSNo significant differences were found in cell growth or measurements between air control and air + hydrogen groups. Compared with air control group, the cells exposed to hyperoxia showed significantly suppressed proliferation, reduced mitochondrial membrane potential, increased MDA content, and decreased SOD activity. Intervention with hydrogen resulted in significantly increased cell proliferation and SOD activity and lowered MDA content, and restored the mitochondrial membrane potential in the cells with hyperoxia exposure (P<0.05).

CONCLUSIONHydrogen can significantly reduce hyperoxia-induced oxidative stress injury in premature rat type II AECs, improve the cellular antioxidant capacity, stabilize the mitochondrial membrane potential, and reduce the inhibitory effect of hyperoxia on cell proliferation.
